Effect of Substrate Type on Survival and Growth of Sweet Potato Vitroplants Under Acclimatization

Abstract: The lack of healthy planting materials is one of the major constraints of sweet potato cultivation in Benin. One of the solutions to this problem is the use of vegetal biotechnology through micropropagation. At the end of the production of vitroplants, a crucial phase consists in acclimatizing them before their transfer to the real environment. The type of substrate used plays an important role in the success of acclimatization. This study aims at evaluating the effect of the type of substrates on the survival and growth of sweet potato vitroplants in acclimatization of the two accessions (Mèché and Bombo vôvô). For this, three substrates were assessed. These are Potting soil alone, Potting soil + Sawdust, and potting soil + Charcoal powder in proportion 2:1. These substrates were first sterilized separately in an oven at 150°C for 2 hours. The experiment was conducted in the greenhouse of Central Laboratory of Vegetal Biotechnology and Plant Improvement of the Faculty of Sciences and Techniques of the University of Abomey-Calavi following a model of random complete blocks in split plot design. The results show that the type of substrate influences the success of acclimatization in terms of survival rate, the gain of size and the number of neoformed leaves (p= 0.0033; 0.0019 and 0.0001). The Potting soil + Charcoal powder substrate influenced these parameters better than the other two with 95%, 3.13 cm and 3.165 respectively for survival rate, size gain and the number of neoformed leaves. It was followed by the Potting soil + Sawdust substrate. Acclimatized plants have been successfully introduced into the field, proving that this technique is fruitful in the increase of the production of healthy cuttings and make them available to farmers. The protocol used in this study can be applied in the acclimatization of sweet potato vitroplants.

Abstract: Barley stripe mosaic virus, the type member of Hordeivirus, constitutes an important seed-transmitted pathogen distributed in almost every barley growing area worldwide. Researches on efficient detection of BSMV suitable in wild application are in urgent need. New platforms like microfluidic technology keep progressing rapidly in past decades and gains increasing influence on life sciences. Thus in this study, one rapid screening method for BSMV via microfluidic chip was established. Through method validity, we succeeded in demonstrating the amplification of BSMV within 30 min at one constant temperature using a microfluidic chip assay. Sensitivity analysis conformed that this method obtained a detection limit at 1.00×102 copies/μL, which was even more sensitive than real-time RT-PCR. Maize chlorotic mottle virus, Maize dwarf mosaic virus, Oat mosaic virus, and Wheat streak mosaic virus were chosen in specificity test along with BSMV, but only BSMV expressed typical amplification curves and Ct. Microfluidic analytic systems realize miniaturization by reducing the reaction volume to just 5 μL, which has several superiorities such as saving expensive reagents and reducing inspection costs. Moreover this microfluidic chip assay can simultaneously detect a variety of viruses, and the whole process takes just 1 hour, which greatly speeds up the detection and improves efficiency. All in all, this microfluidic chip method has the potential to be further implemented by phytosanitary services for routine diagnose as well as rapid screening in places like ports.

Abstract: Developing new durum wheat varieties with high yielding, quality, stress tolerant, uniform and stable is the most important of breeders goal. Benya is commercial name given to a newly released durum wheat variety originated from ICARDA. Twenty durum wheat genotypes were tested under regional variety trial along with two standard checks (Bulala and Dire) and with local check (Ingilize) for two successive years (2019 and 2021) across three environments namely; Sinana, Agarfa and Ginir representing Southeastern Ethiopia. Data analysis of yield showed that, Begna gave highest mean yield up to 4.94t ha-1 compared to nineteenth test entries. Moreover, Begna showed more stable grain yield performance, standard quality, disease tolerant, uniform, high protein and gluten content, high biomass and it is early maturing variety. Farmers and others stakeholders were allowed participatory variety selection and Begna is their preferred variety in the trial. Under variety verification it was evaluated both on farmers’ field and research station along with Tasfaye standard check and Ingilize local check during 2021/2 cropping season. Finally Begna was officially released in 2022 for high and mid altitude of Southeastern Ethiopia. It is commercial variety and offers new hope for farmers in rust-prone regions.

Abstract: Tobacco rattle virus (TRV), member of the genus Tobravirus, is an important plant pathogen with a wide host range beyond any known plant viruses. It is able to be transmitted by nematodes of the genera Trichodorus and Paratrichodorus (Trichodoridae). In this paper, a rapid screening method was established for inspection of TRV in seeds or other plant materials, combining microfluidic chip technique and fluorescence detection system. The particular region (located at 496-995) of TRV strain (accession number: KP100069.1) was adopted for primer design, method establishment and improvement. Through analysis, this method was proved to be specific for only detecting sample infected by TRV, while 7 other leaf materials infected by Tomato spot wilt virus (TSWV), Tobacco ringspot virus (TRSV), Cucumber mosaic virus (CMV), Lily symptomless virus (LSV), Lily mosaic virus (LMV), Tomato ringspot virus (ToRSV), and Prunus necrotic ringspot virus (PNRSV), respectively, showed negative results. Sensitivity tests represented a detection limit as low as 1.00×102 copies/μL which is beyond normal PCR assays. In one word, microfluidic chip based analyzing platforms hold high promises to enable high-throughput and high-precision screening with less sample consumption, fast detection, simple operation, multi-functional integration, small size, multiplex detection and portability, thus promoting the development of biosecurity diagnostics.
