New Nickel Metal Complex towards Biomimetic Catecholase Enzyme Oxidation: Synthesis and Kinetics Studies

Abstract: Pyrano[2,3-c]pyrazole ligand and their transition metal complex with nickel (NiIIMPC) were synthesized and characterization by elemental analyses, magnetic susceptibility, FT IR, UV-Vis, and mass spectroscopy. Complex have been investigated as functional models for catechol oxidase activity by using a catechol as a model substrate. According to the kinetic measurement results, the rate of catechol oxidation follows first order kinetics. Nickel complex has been used in homogenous oxidation of catechol in presence of a green oxidant H2O2. kinetic parameters were obtained Vmax= 2.698×10-3MS-1, Km = 7.878M, kcat= 4.496×10-2S-1 and kcat/Km = 0.5707 M-1S-1.

Evaluation of Biofield Energy Treated Vitamin D3 on Bone Health Parameters in Human Bone Osteosarcoma Cells (MG-63)

Abstract: The study was aimed to evaluate the effect of Consciousness Energy Healing based vitamin D3 and DMEM medium on bone health parameters. The test items, were divided into two parts. One part of each sample received the Consciousness Energy Healing Treatment by Dezi Ann Koster and those samples were labeled as the Biofield Energy Treated (BT) samples, while the other parts of each sample were denoted as the untreated test items (UT). Different parameters were performed for the evaluation of bone health like ALP, collagen, and bone mineralization in human bone osteosarcoma cells (MG-63). The cell viability (MTT) data showed the test samples were found as safe in the tested concentrations. The level of ALP was significantly increased by 431.44% and 436.87% in the BT-DMEM + UT-Test item and BT-DMEM + BT-Test item, respectively at 1 µg/mL compared to the UT-DMEM + UT-Test item group. Further, the ALP level was significantly elevated by 163.34%, 44.91%, and 57.67% in the UT-DMEM + BT-Test item, BT-DMEM + UT-Test item, and BT-DMEM + BT-Test item groups, respectively at 50 µg/mL compared to the untreated. Collagen was significantly increased by 56.58%, 84.95%, and 43.27% in the UT-DMEM + BT-Test item, BT-DMEM + UT-Test item, and BT-DMEM + BT-Test item, respectively at 0.1 µg/mL compared to the untreated. Further, the collagen level was significantly increased by 21.27%, 47.26%, and 63.1% in the UT-DMEM + BT-Test item, BT-DMEM + UT-Test item, and BT-DMEM + BT-Test item groups, respectively at 50 µg/mL compared to the untreated. Besides, the percent of bone mineralization was distinctly increased by 41.87% and 91.87% at 1 µg/mL in the BT-DMEM + UT-Test item and BT-DMEM + BT-Test item groups, respectively while, increased by 72.66% and 186.68% at 50 µg/mL in the UT-DMEM + BT-Test item and BT-DMEM + BT-Test item groups, respectively compared to the untreated. The percent of bone mineralization was distinctly increased by 101.21%, 57.76%, and 125.53% in the UT-DMEM + BT-Test item, BT-DMEM + UT-Test item, and BT-DMEM + BT-Test item, respectively at 10 µg/mL compared to the untreated. Altogether, the Biofield Energy Treated vitamin D3 was significantly improved the bone health parameters and it could be an excellent alternative nutraceutical supplement against various bone-related disorders including osteoporosis, low bone density, osteogenesis imperfecta, Paget’s disease, rickets, osteomalacia, deformed bones, autoimmune and inflammatory diseases, stress management and prevention, and anti-aging by improving overall health.

Abstract: Cellulosic biomass, the most remarkable renewable source of fuels and other compounds of potential importance, needs highly complex mixture of various enzymes for its degradation. For sustainable and economical bioprocess, the availability of these enzymes in high quantities and at a low price is much warranted. The advancements in discovery of new strains, highly valuable techniques and molecular biotechnological tools have led to Pichia pastoris gaining a recognized organism status both at industrial as well as laboratory level. With numerous beneficial characteristics on its credit P. pastoris has emerged a promising host for most of heterologous proteins production. Thus, the production of fungal cellulytic enzymes is of worth to be comprehensively illustrated. There are some activators including promoters used in the expression host to increase the utility of expression system. This review summarizes the heterologous expression of fungal cellulases (Exo-glucanases, endo-glucanases (EGs) and beta-glucosidases (BGLs)) and advocates the various kinds of promoters used in P. Pastoris for expression of these enzymes and proteins. Further, concluding remarks will provide insights revealing the scope of P. pastoris as a potential expression system tool in today's modern direction of research.

One part of each sample received the Consciousness Energy Healing Treatment by Gary Richard Gerber and those samples were labeled as the Biofield Energy Treated (BT) samples, while the other parts of each sample were denoted as the untreated test items (UT). ALP, collagen, and bone mineralization activities were performed to assess bone health in human bone osteosarcoma cells (MG-63). The cell viability assay (MTT) data showed that the test samples were found as safe in all the tested concentrations. The level of ALP was significantly increased by 74.54% and 98.26% in the UT-DMEM + BT-Test item and BT-DMEM + UT-Test item groups, respectively at 10 µg/mL compared to the UT-DMEM + UT-Test item group. Further, the ALP level was significantly elevated by 111.17% in the BT-DMEM + BT-Test item group at 100 µg/mL compared to the UT-DMEM + UT-Test item group. Collagen was significantly increased by 147.46%, 59.67%, and 35.36% in the UT-DMEM + BT-Test item group at 1, 10, and 50 µg/mL, respectively compared to the untreated group. Moreover, the level of collagen was significantly increased by 89.88%, 86.42%, and 82.08% in the UT-DMEM + BT-Test item, BT-DMEM + UT-Test item, and BT-DMEM + BT-Test item groups, respectively at 50 µg/mL with respect to the untreated group. Further, the collagen level was significantly increased by 365.76%, 80.10%, and 76.09% in the BT-DMEM + BT-Test item group at 0.1, 10, and 50 µg/mL compared to the untreated group. Besides, the percent of bone mineralization was distinctly increased by 59.30%, 30.69%, and 177.7% in the UT-DMEM + BT-Test item, BT-DMEM + UT-Test item, and BT-DMEM + BT-Test item groups, respectively at 50 µg/mL compared to the untreated group. The percent of bone mineralization was distinctly increased by 29.44%, and 45.51% in the BT-DMEM + UT-Test item and BT-DMEM + BT-Test item groups, respectively at 100 µg/mL compared to the untreated group. Overall, the Biofield Energy Treated vitamin D3 was remarkably improved the bone health parameters and help to combat vitamin D3 deficiency and fight against various bone related problems including low bone density, osteoporosis, osteogenesis imperfecta, Paget’s disease of bone, rickets, osteomalacia, bone and joint pain, bone fractures, deformed bones, osteoma, chondrodystrophia fetalis, autoimmune and inflammatory diseases, stress management and prevention, and anti-aging by improving overall health.
